The high-speed laser-scanning FRAP photobleaching module uses galvanometers for microscopy, with monochromatic/multicolor lasers via single-mode PM fiber input. Multiple ROIs of different sizes, positions and shapes can be precisely targeted simultaneously for bleaching, localized cell illumination and programmed paths. Applications include photobleaching, photoactivation/photoconversion in biology/materials. Highly integrated, easy to install and compatible with Olympus, Nikon and other commercial microscopes, it studies intracellular protein dynamics economically without point-scanning confocal equipment. Scanning: Precision high-speed XY motors with highly reflective mirrors focus the beam and trace preset vectors, such as rapid spots or area fills, delivering high energy to selected ROIs for immediate bleaching/activation. TTL control enables microsecond switching and dynamic power, limiting illumination to ROIs and reducing off-target phototoxicity. Kayja's dedicated software integrates path planning, pattern drawing, power modulation and camera acquisition. Patterns include points, lines, serpentine fills and custom closed curves. Laser switching synchronizes with scanning and stays off otherwise. Millisecond external hardware triggers accurately send/receive TTL for zero-delay synchronization with sCMOS shutters. Optical/mechanical expansion connects to upright/inverted microscopes through dedicated adapters; fine lens adjustment calibrates wavelength-dependent chromatic aberration and sample focal planes.
